MISSION™ shRNAs Specifically Knock Down Gene Expression in Human and Mouse

imaGenes provides in cooperation with Sigma-Aldrich MISSION™ shRNAs targeted against human and mouse genes established by The RNAi Consortium (TRC).

MISSION™ shRNAs are expressed within the cell from a plasmid and serve as potent gene silencing agents which allow long-term gene knockdown. The plasmid containing the shRNA can be used either directly for transfection to achieve transient gene knockdown or as lentiviral particle for long-term experiments by integration into the host genome.

For a schematic overview of shRNA mediated gene silencing, click here.

Currently, the MISSION™ shRNA collection targets 16068 human and 15808 mouse genes.

Benefits of the shRNA approach

  • Straightforward – avoid handling of RNAs and Dicer treatment
  • Comprehensive – average of 3-5 shRNA constructs per target gene
  • Efficient – high likelihood for successful knockdown with multiple constructs per target
  • Economical – Vector based system provides a renewable resource compared to siRNA
  • Flexible – Transient or stable silencing (puromycin selection) for long term studie more...
  • Solutions – Lentiviral system for transduction of difficult cell lines (non-dividing and primary cells)
  • Quality – All clones are sequence verified

MISSION™ shRNA Product Overview

The MISSION™ shRNA libraries for human and mouse are currently provided as

  • purified plasmid DNA
  • frozen bacterial glycerol stock harboring sequence-verified shRNA.
  • lentiviral transduction particles

Each gene is represented by a target set that consists of an average of 3-5 individual constructs or clones targeting different regions of the gene sequence. Bacterial cultures may be amplified from the glycerol stocks for use in purification of the shRNA plasmid DNA. Alternatively, the purified plasmid format does not require these extra steps as the growth and DNA purification are completed for you. Target cell lines may be transfected with the purified plasmid for transient or stable gene silencing (puromycin selection). more...

MISSION™ shRNA Gene Family Sets

The MISSION™ TRC shRNA gene family sets are collections of genes related to specific functional classes. more ...
